Oxidized polysaccharides for anticancer-drug delivery: What is the role of structure?

Study provides an in-depth analysis of the structure-function relationship polysaccharide anticancer drug carriers and points out benefits potential drawbacks differences in glycosidic bonding, branching binding mode carriers. Cellulose, dextrin, dextran hyaluronic acid have been regioselectively oxidized to respective dicarboxylated derivatives, allowing them directly conjugate cisplatin, while preserving their major structural features intact. The structure source has crucial impact on conjugation effectiveness, carrier capacity, release rates, vitro cytotoxicity cellular uptake. For example, branched dextrin-based partially counter undesirable initial burst release, it also attenuates uptake carried drug. Linear polysaccharides containing β-(1→4) bonds at C2 C3 (cellulose hyaluronate) best overall combination for improved delivery applications including potentiation cisplatin efficacy towards malignances.
